If a teen asking about EC says that she has been raped or sexually abused, give her the hotline number of the local sexual assault center. The center will offer her options, including information about services at a local hospital and a forensic sexual assault exam. The purpose of the present study was to examine the effect of this highly toxic aldehyde on the activity of cytochrome c oxidase COX ; and oxidative modifications of mitochondrial proteins. Further, we also examined whether aging increases formation of HNE modified proteins in hearts from male Wistar rats. Incubation of cardiac mitochondria with HNE resulted in concentrationand time-dependent loss of COX activity. Using immunoprecipitationWestern blot analysis we found that the loss of enzyme activity corresponds with the formation of HNE adducts with COX subunit I. Moreover, HNE treated mitochondria exhibited reduced thiol group content, increased lipid peroxidation and appearance of HNE-lysine adducts and bityrosines. These markers of oxidative damage and COX activity were also measured in hearts from adult 6 mo ; , old 15 mo ; and senescent 26 mo ; rats. The activity of COX decreased progressively during aging. The reduced activity was accompanied by a progressive increase in HNECOX adducts, whereas the relative amounts of COX subunit I were not significantly changed in the aged and senescent rats. Aging was also associated with accumulation of markers of protein and lipid oxidation. These observations indicate that during short term incubation HNE selectively.
